Advanced bioprocess monitoring by implementation of Proton Transfer Reaction - Mass Spectrometry (PTR-MS) for measurement of volatile components in the bioreactor

چکیده

To demonstrate the proper functionality of the inlet system, three identical E. coli fermentations were conducted. The reproducibility of these experiments is shown by comparison of a set of process variables (Table 1, Figure 2). Figure 1: Simplified schematic of the setup. Fermenter off-gas is sampled and analyzed continuously. The inlet box, containing the flowcontrollers, is thermally controlled and allows for dilution and calibration and, if needed, multiplexing to analyze several fermenters (not shown).

Ptr-ms Technology for Process Monitoring and Control in Biotechnology

Process monitoring is becoming increasingly relevant in biotechnology, especially in the manufacturing sector to promote cost-effective production. Proton transfer reaction mass spectrometry (PTR-MS) is a well established analytical tool for the measurement of volatile organic compounds (VOCs) and offers fast, real-time detection and quantification of VOCs at trace concentrations. In addition, ...

PTR-MS in Italy: A Multipurpose Sensor with Applications in Environmental, Agri-Food and Health Science

Proton Transfer Reaction Mass Spectrometry (PTR-MS) has evolved in the last decade as a fast and high sensitivity sensor for the real-time monitoring of volatile compounds. Its applications range from environmental sciences to medical sciences, from food technology to bioprocess monitoring. Italian scientists and institutions participated from the very beginning in fundamental and applied resea...

Advanced off-gas measurement using proton transfer reaction mass spectrometry to predict cell culture parameters

Background Mass spectrometry is a well-known technology to detect O2 and CO2 in the off-gas of cell culture fermentations. In contrast to classical mass spectrometers, the proton transfer reaction mass spectrometer (PTR MS) enables the noninvasive analysis of a broad spectrum of volatile organic compounds (VOCs) in real time. The thereby applied soft ionization technology generates spectra of l...

High-Resolution Measurement of Volatile Organic Compounds Dissolved in Seawater Using Equilibrator Inlet-Proton Transfer Reaction-Mass Spectrometry (EI-PTR-MS)

We have developed an equilibrator inlet-proton transfer reaction-mass spectrometry (EI-PTRMS) system for the high-resolution measurement of the concentrations of multiple volatile organic compounds (VOCs) dissolved in seawater. The equilibration of six VOCs (dimethyl sulfide (DMS), isoprene, propene, acetone, acetaldehyde, and methanol) between seawater samples and the carrier gas, and the resp...

On-line monitoring of microbial volatile metabolites by proton transfer reaction-mass spectrometry.

A method for analysis of volatile organic compounds (VOCs) from microbial cultures was established using proton transfer reaction-mass spectrometry (PTR-MS). A newly developed sampling system was coupled to a PTR-MS instrument to allow on-line monitoring of VOCs in the dynamic headspaces of microbial cultures. The novel PTR-MS method was evaluated for four reference organisms: Escherichia coli,...
